Abstract

Latar belakang
Insiden astrositoma di RSUD Dr. Soetomo Surabaya meningkat dalam tiga tahun terakhir. Keterlibatan berbagai growth factor dan reseptornya serta penanda proliferasi dalam patogenesis molekuler neoplasma astrocytic banyak diteliti secara ekstensif. Epidermal growth factor receptor (EGFR) dan Ki-67 terlibat dalam transduksi sinyal mitogenik dan jalur proliferasi sel. Tujuan penelitian ini menganalisis ekspresi EGFR dan Ki-67 pada astrositoma derajat II,III, dan IV, serta menganalisis korelasi antara ekspresi EGFR dan Ki-67 pada derajat histopatologi astrositoma.
Metode
Rancangan penelitian yang digunakan adalah cross sectional. Sampel dibagi dalam; 6 astrositoma derajat II, 7 astrositoma derajat III, and 13 astrositoma derajat IV yang telah didiagnosis selama periode 2009-2013. Sampel dilakukan pulasan immunohistokimia dengan antibodi monoklonal EGFR dan Ki-67. Perbedaan ekspresi EGFR dan Ki-67 pada berbagai derajat astrositoma dianalisa menggunakan uji Kruskal-Wallis. Hubungan antara ekspresi EGFR dan Ki-67 pada astrositoma dianalisa menggunakan uji Spearman.
Hasil
Hasil analisa statistik ekspresi EGFR pada astrositoma derajat II, III, dan IV menunjukkan perbedaan bermakna (nilai p=<0,05). Hasil analisa statistik ekspresi Ki-67 pada astrositoma derajat II, III, dan IV menunjukkan perbedaan bermakna (p<0,05). Hasil analisa statistik ekspresi EGFR dan Ki-67 pada astrositoma menunjukkan perbedaan bermakna (p<0,05).
Kesimpulan
Ekspresi EGFR dan Ki-67 dapat digunakan untuk membedakan diagnostik dan prediktif pada derajat histopatologi astrositoma. Ekspresi EGFR dan Ki-67 pada astrositoma menunjukkan korelasi.

ABSTRACT
Background
The incident of astrocytoma in Dr. Soetomo General Hospital Surabaya have been increased for the last three years. The involvement of various growth factors, growth factor receptors and proliferative markers in the molecular pathogenesis of astrocytic neoplasms are being studied extensively. Epidermal Growth Factor Receptor (EGFR) and Ki-67 is involved in mitogenic signal transduction and cell proliferation pathways. This study was to analyze expression of EGFR and Ki-67 in grade II, III, and IV astrocytomas, as well as analyzing the correlation between EGFR and Ki-67 expressions with histopathological grading in astrocytoma.
Methods
Study design was cross sectional. Sample were divided into: 6 grade II, 7 grade III, and 13 grade IV astro-cytoma were diagnosed in period between 2009-2013. Sample were stained with immunohistochemistry using monoclonal antibody of EGFR and Ki-67. The differences of EGFR and Ki-67 expression on astrocytoma were analyzed by Kruskall-Wallis test. The correlation between EGFR and Ki-67 expresssion were analyzed by Spearman test.
Results
Statistic analyses of EGFR expression on astrocytoma grade II, III, and IV showed significant differences (p<0.05). Statistic analyses of Ki-67 expression on astrocytoma grade II, III, and IV showed significant differences (p<0.05). Statistic analyses of EGFR and Ki-67 expression in astrocytoma (p<0.05).
Conclusion
EGFR and Ki-67 expression may play a role in distinguishing in diagnostic and predictive grading astrocytomas. Expression of EGFR and Ki-67 in grading astrocytomas showed correlation.
